چکیده

Abstract Objectives Endoplasmic reticulum (ER) stress plays a significant role in the progression of diabetic kidney disease (DKD), and dipeptidyl peptidase-4 (DPP4) inhibitors are widely used antihyperglycemic agents, exerting renal beneficial effects DKD. Here, we investigated DPP4 inhibitor Sitagliptin (Sita) ER homeostasis kidneys DBA2/J (D2) mice albumin-stimulated HK-2 cells. Methods Results was observed both vivo vitro , as reflected by notably increased glucose-regulated protein 78 kDa (GRP78), CHOP, high phosphorylation PERK (p-PERK), cleaved caspase3 (c-CASP3), whereas Sita effectively attenuated these disorders. Meanwhile, expression SIRT1 . To further validate potential regulating stress, regulated siRNA overexpressed plasmids albumin-overloaded Elevated alleviated albumin-induced while decreased aggravated albumin-treated Conclusion The results suggest that novel mechanism links enzyme to during tubular injury DKD highlight may be target for managing

Bushen-Yizhi formula ameliorates cognitive dysfunction through SIRT1/ER stress pathway in SAMP8 mice

The Chinese formula Bushen-Yizhi (BSYZ) has been reported to ameliorate cognitive dysfunction. However the mechanism is still unclear. In this study, we employ an aging model, SAMP8 mice, to explore whether BSYZ could protect dementia through SIRT1/endoplasmic reticulum (ER) stress pathway. Morris water maze and the fearing condition test results show that oral administration of BSYZ (1.46 g/kg...

The Role of SIRT1 in Diabetic Kidney Disease

Sirtuins (SIRTs) are members of the silent information regulator 2 family. In mammals, of the seven known SIRTs, SIRT1 function is most studied and has been shown to regulate wide range of cellular functions that affect metabolic homeostasis and aging. SIRT1 exerts anti-apoptotic, anti-oxidative, and anti-inflammatory effects against cellular injury, and protects the cells through the regulatio...

PGC-1α ameliorates kidney fibrosis in mice with diabetic kidney disease through an antioxidative mechanism

The production of reactive oxygen species (ROS) is a common phenomenon in podocyte impairment, which leads to the irreversible progression of chronic kidney diseases, such as diabetic kidney disease (DKD). Previous research has indicated that peroxisome proliferator‑activated receptor γ (PPARγ) coactivator‑1α (PGC‑1α) participates in mitochondrial biogenesis and energy metabolism in certain mit...

Non-albuminuric Diabetic Kidney Disease in Diabetic Patients: A Review

Introduction: The constantly increasing incidence of type 2 diabetes, probably due to obesity and sedentary life, has led to the increased incidence of macro- and microvascular diabetic complications such as nephropathy. Despite the prompt efforts to develop effective treatments for diabetes and slow the progression of its complications, it is still reported as the most common cause of chronic ...
